Time course of labeling of an inside-out excised patch from Xenopus oocytes by R18 added to the bath imaged with confocal microscopy. R18 was applied at a concentration above its critical micellar concentration, so that self-quenching reduced its fluorescence in the bath and it became bright only as it partitioned into the cell membrane on the tip of the patch pipette.
