Fluorescent vancomycin does not access the intracellular MRSA reservoir. MRSA (MW2-GFP, green) inside KCs (F4/80, purple) did not get stained by infused fluorescent vancomycin (50 µg, green). Binding of fluorescent vancomycin to MRSA was only observed when unlabeled MRSA (MW2) was injected. These staphylococci appeared in green and were rapidly taken up by KCs. MRSA-AF555 was injected 15 min before initiation of SD-IVM. Fluorescent vancomycin was injected 1 m after initiation of 10-min time interval, followed by injection of unlabeled MRSA for 10 min visualized by SD-IVM. Bar, 25 µm.
