RAW 264.7 macrophages were transfected with the previously characterized p40phox PX-EGFP fusion construct. p40phox PX binds exclusively to PI3P on membranes. Imaging was initiated at the time of phagocytosis of Texas Red-labeled latex beads using an UltraView confocal microscope. W7 was added 11 min after the start of the sequence, demarcated at frame 10. Confocal images were acquired at 37°C at 1-min intervals for 1 h and 15 min. Individual frames are projections of six confocal Z-sections.
