Dorsal view of a pupa that carries hand-GFP and org-1-RFP and in which RNAi against piwi was induced ( org-1>>UAS-dsRNA-piwi). The video starts at late stage P3 and shows that down-regulation of piwi has no impact on the dedifferentiation of the AMs into mononucleate myoblasts but that transdifferentiation is arrested under these conditions after AM dedifferentiation. This suggests an essential role of the Yki target piwi during the lineage reprogramming of the AM derived myoblasts into the founders of the VLM during transdifferentiation.
