Live-cell imaging of the images shown in Fig. 4 A (top). HEK293T cells stably expressing SNAP-macroH2A and the Fucci cell cycle reporter were labeled as described in Fig. 4 A. Live-cell images were acquired every 15 min for 24 h using widefield microscopy. The video has 97 frames total, shown at 8 frames per second. For the Fucci visualization, cells expressed mCherry (red) during the S to G2 phase and TagBFP (blue) during the G1 phase. For the SNAP-tagging visualization, preexisting and newly incorporated macroH2A were labeled with Oregon Green (green) and JF646 (magenta), respectively.
