A PGALSWE1 cell grown in galactose to overexpress SWE1. The mother cell formed two elongated buds. Septin structures at the bud neck (monitored with Cdc3-GFP) became dim as additional septin structures formed in the bud. The cell did not undergo anaphase (monitored with mCherry-Tub1) for the entire imaging time (420 min). TIFF time-lapse images (see Materials and methods for image acquisition details) were converted to AVI format using ImageJ with a compilation of 4.0 frames/second. The AVI file was then converted to MP4 format using the Video Converter Any Format application in Cidade Apps.
