Plk4 is required for acentriolar spindle assembly. Oocytes expressing EGFP-EB3 (green, right; inverted black, left) and histone H2B-mRFP (red, right). Progression through the early stages of meiosis was imaged under the indicated conditions. Treatment with centrinone or coexpression of Plk4Δkinase significantly delays microtubule growth. Inhibition of Plk4 with 5 µM centrinone can be partially rescued by expression of a drug-resistant Plk4 allele (G95R). Z stacks are composed of 20 optical sections every 3 µm. Images were acquired every 15 min. NEBD corresponds to time 00:00 (hours:minutes). See also Fig. 1 (A–E).
