3D rendering of kinetochore expansion in nocodazole-treated extract. Rotating rendering of mitotic chromosomes (blue) assembled in Xenopus egg extracts treated with nocodazole and stained for BubR1 (magenta) to demonstrate the fibrous kinetochore expansion, which surrounds pairs of CENP-A foci (green, sister centromeres), extends out typically along the surface of the chromosome mass, and often overlaps with kinetochores from other chromosomes. One rotation of the whole chromosome mass is shown followed by a zoomed-in rotation focusing on a single chromosome. Chromosome staining is not shown for the second rotation. Bar, 0.5–2 µm, depending on zoom, as indicated. Data were acquired using a DeltaVision OMX V4/Blaze 3D-SIM super-resolution microscope system (GE Healthcare) and the 3D rendering was generated using Imaris software (Bitplane).
