Control mitosis (scrambled LNA). This movie shows that control S2 cells expressing H2B-GFP (green) and mCherry-tubulin (red) that have been treated with scrambled LNA gapmers divide normally. The condensed chromosomes align at the metaphase plate, and sister chromatids are quickly pulled toward opposite poles in anaphase after chromosome alignment is complete. The movie was acquired by time-lapse microscopy using a wide-field fluorescence microscope (DeltaVision Core system; Applied Precision). Frames were taken every 2 min until the end of cell division. Stills are shown in Fig. 2 C. Bar, 15 µm.
