GFP-Arp1A is recruited to sites of ablation and moves processively poleward. Time-lapse spinning disk confocal movie of a metaphase PtK2 cell transfected with GFP-Arp1A (grayscale), which is recruited and moves processively poleward after laser ablation (X; time 00:00 for ablation event in Fig. 5 A and time 01:33 for later ablation event). Arrowheads highlight the movement of GFP-Arp1A puncta after ablation. Note that Arp1A also localizes to kinetochores at mitosis. Time in min:s. Video was collected using a spinning disk confocal microscope (CSU-X1 with Eclipse Ti-E) at a rate of 1 frame every 14 or 7 s and is played back at a rate of 30× real time (see Materials and methods). Video corresponds to the still images from Fig. 5 A and one additional ablation event.
