Time-lapse imaging of SNAP-TMR-Star–labeled SNAP-uLCa in HeLa clone 3.3 cells transiently transfected with GFP-pericentrin reveals clathrin moving toward centrosomes in a G2/M-transitioning cell. Asynchronous clone 3.3 cells were transiently transfected with GFP-pericentrin (green) 48 h before labeling SNAP-uLCa with SNAP-TMR-Star (red). Cells were kept in an incubation chamber (37°C in 5% CO2 at 95% humidity) during imaging by time-lapse confocal microscopy (DM1 6000 CS, SP5). 2D images were acquired over a 2-min period with frames taken every 10 s.
