Time-lapse imaging of an actin focus in a wild-type embryo expressing GFP-actin with the mesodermal driver twi-GAL4. Time-lapse sequence from a live stage 14 wild-type embryo expressing GFP-actin in all muscle cells with twi-GAL4. Images were acquired using a laser-scanning confocal microscope (LSM 700; Carl Zeiss). A single optical slice of a confocal stack is shown. 28 frames were taken at 20-s intervals. Total sampling length was ∼9 min. Note that the actin-enriched focus maintained a “dense core” throughout its lifespan.
