Time-lapse movie of negative-contrast fluorescence micrographs of an adf1-M2 cell expressing Rlc1p-3GFP. Images are maximum intensity projections (ImageJ) of Z-series of confocal stacks at each time point, acquired with a microscope (model IX-71; Olympus) equipped with a spinning disk-confocal unit (Ultra-view RS; PerkinElmer) and an Orca-ER camera (Hamamatsu Photonics). Bundles of actin filaments grew from two clumps of nodes in the middle of the cell and merged together into a ring that then constricted (Fig. 6 D shows the same cell). Time in minutes.
