Time-lapse movie of negative-contrast fluorescence micrographs of an adf1-M3 cell expressing Rlc1p-TdTomato (red) and GFP-CHD (green). Images at each time point are maxim intensity projections (ImageJ) of three confocal slices from the top of a dividing cell, acquired with a microscope (model IX-71; Olympus) equipped with a spinning-disk confocal unit (model CSU X-1; Yokogawa) and an EMCCD camera (Ixon; Andor). Actin patches tagged with GFP-CHD concentrated at both ends of the cell and persisted far longer than the normal 20 s in wild-type cells. At the start of the movie, nodes marked with Myo2 have aggregated into three clumps (arrows) visible in these three sections. Over time, bundles of actin filaments grew from these clumps of nodes and merged together into a ring at the end of the movie. Time in seconds.
