Time-lapse movie of negative-contrast fluorescence micrographs of an adf1-M3 cell expressing Rlc1p-3GFP at 25°C. Images are single confocal sections focused on the top section of the dividing cell at each time point, acquired with a microscope (model IX-71; Olympus) equipped with a spinning-disk confocal unit (Ultra-view RS; PerkinElmer) and an Orca-ER camera (Hamamatsu Photonics). Nodes appeared and moved together into three large clumps. Time in seconds.
