Time-lapse movie of negative-contrast fluorescence micrographs of a wild-type cell expressing Rlc1p-3GFP at 25°C. Images are single confocal sections focused on the top section of the dividing cell at each time point, acquired with a microscope (model IX-71; Olympus) equipped with a spinning-disk confocal unit (Ultra-view RS; PerkinElmer) and an Orca-ER camera (Hamamatsu Photonics). The broad band of nodes coalesced into a contractile ring (Fig. 4 A shows the same cell). The cell is circled in the first frame. Time in seconds.
