Time-lapse movies of fluorescence micrographs of an adf1-M2 cell expressing Rlc1p-3GFP and SPB marker Sad1p-mEGFP at 25°C. Images are maximum intensity projections (ImageJ) of Z-series of confocal stacks at each time point, acquired with a microscope (model IX-71; Olympus) equipped with a spinning-disk confocal unit (model CSU X-1; Yokogawa) and an EMCCD camera (Ixon; Andor). The cell initially failed to assemble a contractile ring from nodes between time 0 and 22 min but recovered by bundling Myo2 and actin filaments into a ring by 84 min, which then constricted at 92 min. Time in minutes.


Actin filament severing by cofilin is more important for assembly than constriction of the cytokinetic contractile ring
