Time-lapse movie of fluorescence micrographs of an adf1+ cell expressing Rlc1p-3GFP and a SPB marker Sad1p-mEGFP at 25°C. Images are maximum intensity projections (ImageJ) of Z-series of confocal stacks at each time point, acquired with a microscope (model IX-71; Olympus) equipped with a spinning-disk confocal unit (model CSU X-1; Yokogawa) and an EMCCD camera (Ixon; Andor). Nodes coalesced into a ring during 12 min after the separation of SPBs and began to constrict around 35 min later. Time in minutes.
