Speckle dynamics during heat shock. Speckle dynamics were monitored by live- cell imaging of the CHO DG44 cell line (Hsp70_5_P9_4), stably expressing EGFP-lac repressor (green) and mRFP-ASF/SF2 (red). The video shows enlargement of a pre-existing speckle (red) together with decondensation and increased association of encircling BAC transgenes (green) around the speckle exterior. Live cells were maintained at 37°C in a closed chamber system (FCS2; Bioptechs) and were imaged with a 63x objective (NA 1.40) using a Zeiss Axiovert 100M microscope equipped with a CoolSNAP HQ (Roper Scientific) slow-scan CCD camera. Display is at 2 frames/second.
