Movement of Hsp70 BAC transgenes during heat shock. Chromosome movement was monitored by live-cell imaging of the CHO DG44 cell line (Hsp70_5_P9_4), stably expressing EGFP-lac repressor (green) and mRFP-ASF/SF2 (red). Long-range movement of Hsp70 BAC transgenes (green) towards a ASF/SF2 speckle (red) occurred within a 5-min heat shock period. Live cells were maintained at 37°C in a closed chamber system (FCS2; Bioptechs) and were imaged with a 63x objective (NA 1.40) using a Zeiss Axiovert 100M microscope equipped with a CoolSNAP HQ (Roper Scientific) slow-scan CCD camera. Display is at 2 frames/second.
