CYCA RNAi. This video shows S2 cells expressing H2B-GFP and mCherry-tubulin undergoing mitosis after CYCA depletion. Cells show severe mitotic defects, including defective spindles and missegregation of chromosomes. Most cells stop dividing after 2–3 d of RNAi treatment, and many do not progress beyond prometaphase (this video shows day 2 after RNAi treatment). The cytokinesis defect giving rise to multinucleated cells in this video is the most distinct phenotype compared with CID, CENP-C, or CAL1 depletion. Stills are shown in Fig. 2. Bar, 15 µm.
