Expression of the WT(ΔH) cofilin plasmid rescues the suppression of the EGF-induced F-actin assembly and protrusion observed in the cofilin knockdown cells. MTLn3 cells were treated with the cofilin siRNA for 4 h. 2 h after termination of the transfection, they were platedon glass-bottom dishes. When cells attached (~ 8 h), they were transfected with the WT(ΔH)cofilin plasmid for 45 min. After 17 h, the cells were starved for 3–4 h, and then EGF (5 nM) was added to the media (bath application) and cells were imaged for 10 min, after addition of EGF, at a rate of 6 frames/min. At the time of imaging, cells were at 36 h after the siRNA treatment.
