Time-lapse video of the blockade of EC tube morphogenesis by the MMP inhibitor GM6001 in 3D collagen matrices. Transmitted light time-lapse images of several HUVECs in a 3D type I collagen gel cultured in the presence of 5 µM GM6001, which is a broad-spectrum chemical metalloproteinase inhibitor. ECs are unable to under EC tubular network formation in the presence of GM6001 and instead send out fine cell processes like that seen after TIMP-2 or -3 addition. The time lapse covers a period from 2 to 47 h of culture (45 h total). Frames were collected every 30 min and are displayed at 0.1667 frames/s.
