Localization of early CD4+ and CD8+ T cell activation in draining LNs. Extended visualization of data presented in Fig. 3. ∼3 × 106 CMTMR- and CMFDA-labeled OT-I and OT-II T cells were transferred into recipient animals, which were 1 d later immunized with 10 µg OVA, along with Lyve-1 antibody conjugated to quantum dot 705 for labeling the LS. After 6 h, draining LNs were isolated, fixed, and visualized via two-photon serial section microscopy for the location of T cell clusters. B cell, T cell, and lymphatic zones are denoted for visual clarity.
