Islet hyperplasia and preferential localization of glucagon- and somatostatin-producing cells close to adjacent ducts. Pancreata ( n = 3) from 2-mo-old Sst-Cre::Pax4-OE animals (bottom) or matched controls (top) were sectioned (8 µm thickness). 25 consecutives sections were stained with fluorescent DBA (blue), antisomatostatin (red), and antiglucagon (green). Representative islets of Langerhans were photographed on each section, and the islet contour was manually laid out according to DAPI counterstaining. Using Amira software (Mercury), the pictures were aligned to form a z-stack, and movies of the resulting isosurface representation were generated. Note the preferential localization of non–β cells close to ducts in 2-mo-old transgenics as compared with controls.
