SAT III knockdown mitosis. This movie shows that SAT III–depleted S2 cells expressing H2B-GFP (green) and mCherry-tubulin (red) display chromosome segregation defects. The chromosomes condense and align at the metaphase plate normally. Most sister chromatids are pulled toward opposite poles in anaphase. However, some chromosomes are lagging and eventually form micronuclei. The movie was acquired by time-lapse microscopy using a wide-field fluorescence microscope (DeltaVision Core system; Applied Precision). Frames were taken every 2 min until the end of cell division.
